چکیده
مقدمه: جنگلهای هیرکانی، زیستگاهی با رویش باستانی از درختان پهنبرگ هستند که در امتداد دامنههای شمالی رشتهکوه البرز و نزدیک مرزهای جنوبی دریای خزر گسترش یافتهاند. امروزه، فعالیتهای انسانی تأثیر بهسزایی در کاهش پوشش جنگلی داشته است. علاوه بر این شناسایی تنوع گونهای، ساختار جنگل و دستکاریهای انسانی در جنگلهای هیرکانی کمتر مورد توجه قرار گرفته است. بیمهرگان بزرگ جزء اصلی تنوعزیستی خاک در جنگلها هستند. کرمهای خاکی یکی از اجزای اصلی تنوعزیستی خاک جنگلها بوده و یکی از با ارزشترین شاخصهای سلامت خاک و جزء حیاتی فون خاک هستند. با این حال، مطالعه آنها بر اساس ویژگیهای ریختی چالشهای زیادی را به همراه دارد. بارکدینگ DNA یک روش مفید است که به طور گسترده برای مطالعه تنوعزیستی استفاده شده است. بنابراین، هدف از این مطالعه استفاده از رویکردهای مولکولی برای شناسایی کرمهای خاکی و تنوع آنها و مقایسه جوامع این جانوران بین جنگلهای طبیعی و دستکاشت در منطقه هیرکانی است.
مواد و روشها: این مطالعه در بخش مرکزی جنگل هیرکانی و در سه منطقه، انجام شده است. به طور کلی، در هر منطقه، دو ناحیه دستکاشت و دو ناحیه طبیعی انتخاب شده است. در هر ناحیه، شش کودرات به صورت تصادفی در محیط قرار گرفتهاند. به منظور، مطالعات ژنتیکی و نگهداری طولانی مدت، نمونهها در اتانول 96 درصد نگهداری شدند. سپس نمونهها براساس ویژگیهای ریختشناسی، طبقهبندی شدند. در مجموع در این مطالعه، 251 نمونه جمعآوری شده و 22 فرد برای مطالعات ژنتیکی انتخاب شده است. مطالعات مولکولی، بااستفاده از ژن سیتوکروم c اکسیداز زیر واحد یک (COI) انجام شد. سپس آنالیزهای آماری تک متغییره و چند متغییره برای بررسی تنوع کرمهای خاکی و مقایسه جوامع آنها، بین ناحیه دست کاشت و طبیعی انجام شدند.
نتایج: براساس مطالعات ژنتیکی، 15 واحد تاکسونومیک در این بررسی شناسایی شدهاند. یافتههای این پژوهش بیانگر این است که تعداد و تنوع OTUها در نواحی طبیعی و دستکاشت تقریباً یکسان است. نتایج آنالیزهای آماری تکمتغیره و آنالیز چندمتغیره، تفاوت معنیداری را برای کرمهای خاکی بین دو ناحیه طبیعی و دستکاشت نشان نداد.
بحث: این مطالعه نشان میدهد که استفاده از بارکدینگ DNA نتایج دقیقتری را ارائه میدهد، اما برای طبقهبندی دقیقتر نیاز به یک کتابخانه جامع مرجع DNA وجود دارد. با توجه به عدم معناداری تفاوت کرمهای خاکی، بین جنگلهای طبیعی و دستکاشت و معناداری این تفاوت بین سه منطقه مورد مطالعه، اینطور نتیجهگیری میشود که وابستگی کرمهای خاکی به جنگلهای دستکاشت بسته به ویژگیهای خاص جنگلهای دست کاشت، شرایط خاک و شیوههای مدیریتی میتواند متفاوت باشد. علاوه بر این، این نتیجه میتواند نشان دهندهی آن باشد که کرمهای خاکی توانایی بالایی در سازگاری با طیف وسیعی از محیطها دارند. بهطورکلی پیشبینی میشود که در جنگلهای ثانویه که بیش از 20 سال کاشته شدهاند، جنگل زمان کافی برای ترمیم جمعیتهای خود، بهویژه کرمهای خاکی، داشته باشد. بهطور کلی، مطالعهی فعالیتها و تعاملات بین کرمهای خاکی و جنگلهای دستکاشت میتواند دانش ما را درباره سلامت خاک، چرخه مواد مغذی و عملکرد اکوسیستم در این مناطق افزایش دهد و زمینه را برای مدیریت صحیح اکوسیستمهای جنگلی، بهویژه جنگلهای هیرکانی که از قدمت و اهمیت بالایی برخوردار هستند، قراهم آورد.
